This is all your opinion, so obviously there is no right answer.
It would be better if it was from a Movie, but I guess TV and theater count.
Here's a few of mine:
Bill Murray, Lost in Translation
Christian Bale, American Psycho
Daniel Day-Lewis, There will be Blood
Heath Ledger, The Dark Knight
Tom Hanks, Castaway
Robin Williams, Good Will Hunting
and my wild card:
Robert Downey Jr, Tropic Thunder
best acting job by non-traditional actor:
Meatloaf, Fight Club _________________ ♪ ♫One good thing about music, when it hits, you feel no pain...

I watched the Dark Knight for the first time a month or two ago (yes, I've been living in a cave) and was so impressed with Ledger's joker portrayal, I made it my avatar.

DeNiro's performances in Raging Bull and Taxi Driver leap to mind. That's when he was at the peak of his powers. It's so hard to see him in Focker mode now.
Maria Falconetti's performance in The Passion of Joan of Arc is often hailed as the best acting performance ever and it's certainly a great one. If there's a case to be made that acting is all in the eyes then that performance is your thesis statement.
Also, Martin Sheen is incredible in Badlands and Apocalypse Now. _________________ 14-5-3-12

Recently, this one really struck me, Charlie Chaplin in the Great Dictator. Consider that he made this movie before we entered the war, and it was his first talking picture.
Mark Hamill playing the voice of The Joker. When I went to a convention long time ago, I saw Mark Hamill and did The Joker voice. Still amazes he did the voice of it. _________________ (❍ᴥ❍ʋ) ʕʘᴥʘʔ (⌐ ͡■ ͜ʖ ͡■) (┛◉Д◉)┛( ͡° ͜ʖ ͡°) ༼;´༎ຶ ༎ຶ༽
